AOD-9604: A Peptide Fragment Derived from Growth Hormone

AOD-9604 is a synthetic fragment of human growth hormone investigated in metabolic research, and this article reviews the limits of the evidence.

AOD-9604 is a synthetic 16-amino-acid peptide derived from the C-terminal fragment of human growth hormone (residues 177-191). Its development was driven by a specific research question: could the lipolytic signal of hGH be separated from its metabolic effects, particularly the hyperglycaemia and insulin resistance associated with direct GH administration? Studies reported that it did not measurably affect IGF-1 or insulin sensitivity, although its effect on body fat in larger human trials was limited.

Mechanism of Action

Laboratory research suggests AOD-9604 acts on lipolysis, the breakdown of stored triglycerides in adipocytes, through a mechanism thought to involve beta-3 adrenergic receptor activation and cyclic AMP elevation. These pathways are distinct from the classical GH receptor pathway. Preclinical studies reported reductions in adipose tissue in animal models, but this has not led to a registered human indication.

What Research Has Examined

Research has examined AOD-9604 mainly in the context of visceral and subcutaneous fat, usually alongside caloric management and exercise. It is not a replacement for lifestyle modification, and human trial results have been modest. Preliminary research has also looked at osteoarthritis and cartilage repair models.

  • Derived from the hGH C-terminal fragment and studied for lipolytic signalling without IGF-1 effects
  • Proposed beta-3 adrenergic pathway activity in adipocytes
  • Studied alongside structured nutrition and exercise rather than as a standalone approach
  • Preliminary signal in joint and cartilage research
