Semax is a synthetic analogue of the adrenocorticotropic hormone (ACTH) fragment 4-7, developed in Russia in the 1980s and registered there for neurological indications. Its primary studied effects relate to cognitive function, neuroprotection, and the regulation of brain-derived neurotrophic factor, which is why it attracts research interest in relation to mental performance and neurological health.
Mechanism of Action
Semax has been studied for its effects on BDNF (brain-derived neurotrophic factor) expression, which preclinical work suggests are pronounced. BDNF is the primary growth factor for neurons: it supports the survival of existing neurons, the growth of new connections, and plays a central role in learning, memory, and mood regulation. Semax has also been examined for effects on dopaminergic and serotonergic systems, and anti-inflammatory effects in the central nervous system have been reported in preclinical studies.
Clinical Applications
In Russia, Semax is registered for use in ischaemic stroke, optic nerve disease, and peptic ulcer disease. Outside Russia, research interest has focused on attention, working memory and neuroprotection, although evidence in healthy adults remains limited. In studies it has been administered as a nasal spray, with the intranasal route providing direct access to the central nervous system via olfactory transport.
