Sermorelin: The Original GHRH Analogue Explained

Sermorelin is a 29-amino-acid analogue of growth-hormone-releasing hormone, and this article reviews its molecular features and clinical research context.

By UAE Peptide Clinic Research Desk

Sermorelin is a 29-amino-acid analogue of growth-hormone-releasing hormone (GHRH), specifically the first 29 amino acids of the 44-amino-acid native hormone. It was the first GHRH analogue registered in the United States (for the assessment of GH deficiency in children) and remains one of the most clinically established compounds in the GH-axis peptide family. Unlike synthetic GH itself, Sermorelin acts through the body's own pituitary pathway, stimulating endogenous GH release rather than replacing it.

Mechanism of Action

Sermorelin binds to the GHRH receptor on pituitary somatotrophs and stimulates the transcription and release of endogenous growth hormone. Because it acts through the natural feedback loop, including somatostatin counter-regulation, it is considered less likely to produce GH excess than direct GH administration. The pulse it generates is physiological in character: brief, appropriately sized, and subject to the normal inhibitory controls that limit overproduction.

Clinical Applications

Sermorelin has been studied in the context of age-related somatotropic decline, the gradual reduction in GH pulsatility and IGF-1 that occurs from the third decade of life. It is sometimes combined with Ipamorelin, which adds a complementary ghrelin-receptor stimulus. Protocol length and dosing are set by the prescribing physician; published protocols have often run for several months.
