The combination of Tesamorelin, Modified GRF 1-29, and Ipamorelin brings together three structurally distinct compounds acting through two separate receptor families, the GHRH receptor and the ghrelin receptor. The mechanistic rationale is that combined stimulation may produce a larger GH response than any single agent, although human data on this specific combination is limited.
The Three-Way Mechanism
Tesamorelin, a stabilised GHRH analogue, provides the primary pulse amplitude; its stability means it contributes a sustained GHRH signal across the dosing interval. Modified GRF 1-29 (CJC-1295 without DAC) is a shorter-acting GHRH analogue that, when co-administered, is thought to sharpen the peak of the immediate pulse. Ipamorelin adds a ghrelin-receptor stimulus that is mechanistically independent of both GHRH analogues, acting through the GHS-R pathway. The proposed result is a physiologically structured pulse with both high amplitude and appropriate duration.
