CJC-1295 and Ipamorelin: The Combination Explained

CJC-1295 and Ipamorelin are examined in growth-hormone-axis research, and this article reviews their receptor mechanisms and combination evidence limits.

The combination of CJC-1295 and Ipamorelin is one of the most discussed GH-axis combinations in physician-supervised longevity medicine. It brings together a long-acting GHRH analogue and a selective ghrelin receptor agonist, two different inputs into the growth hormone axis that research has examined together.

How the Combination Is Thought to Act

CJC-1295 is a GHRH analogue with a drug affinity complex (DAC) modification that extends its half-life significantly, giving a sustained GHRH signal over several days. Ipamorelin is a ghrelin receptor agonist that, in studies, showed little effect on cortisol. GHRH and ghrelin-receptor signalling converge in the pituitary through different second-messenger pathways, and studies giving both types of compound together have reported larger GH pulses than either alone.

What Research Has Looked At

Interest in the combination has centred on body composition, sleep architecture, recovery from training and skin, but there are no large controlled trials of this specific combination for these outcomes. GH secretagogues raise IGF-1, which is why the prescribing physician monitors IGF-1 and sets any dosing to keep it within a physiological range.

  • Formats a physician may prescribe: vial, pen or nasal spray
  • Duration is set by the prescribing physician
  • Evening timing is often discussed because of the natural nocturnal GH pulse
  • IGF-1 monitoring by the prescribing physician during any protocol
